IGFBP3 Activators

Common IGFBP3 Activators include, but are not limited to Retinoic Acid, all trans CAS 302-79-4, Cholecalciferol CAS 67-97-0, Dexamethasone CAS 50-02-2, β-Estradiol CAS 50-28-2 and Genistein CAS 446-72-0.

IGFBP3 activators are compounds that stimulate the expression or activity of IGFBP3 (Insulin-like Growth Factor Binding Protein 3). IGFBP3 is one of the six binding proteins that specifically bind to insulin-like growth factors (IGFs), namely IGF-1 and IGF-2, modulating their activity. These binding proteins play crucial roles in controlling the distribution, function, and concentration of IGFs in various tissues. IGFBP3, in particular, is the most abundant of the IGF binding proteins in the bloodstream and possesses unique attributes, including the capacity to carry more than 90% of circulating IGF-1, the main effector of growth hormone action.

The list of compounds known as IGFBP3 activators is diverse, reflecting the multifaceted regulation of IGFBP3 expression and activity. Compounds such as retinoic acid and vitamin D3 exert their effects by binding to their respective nuclear receptors, subsequently influencing gene transcription processes that can upregulate IGFBP3. Phytochemicals like genistein, a phytoestrogen, and sulforaphane, found in cruciferous vegetables, have also been shown to enhance IGFBP3 expression. The mechanism of action of these compounds often intersects with key cellular signaling pathways. For instance, curcumin, a compound found in turmeric, affects pathways like NF-kB and MAPK, which have downstream implications for IGFBP3. Other compounds, such as 5-aza-2'-deoxycytidine and trichostatin A, target epigenetic mechanisms, underscoring the importance of chromatin structure and DNA methylation in regulating IGFBP3 expression. The diverse range of IGFBP3 activators and their varied mechanisms of action highlight the intricate web of regulatory processes that control the expression and function of this key protein in the IGF system.
