I2PP2A Activators

I2PP2A activators belong to a class of chemicals that are designed to interact with a specific phosphatase enzyme known as I2PP2A, also referred to as SET. This enzyme plays a critical role in the modulation of phosphatase activity within cells. Phosphatases are enzymes that remove phosphate groups from proteins and other molecules, a process which is vital for numerous cellular functions, including cell cycle progression, signal transduction, and the regulation of various metabolic pathways. I2PP2A itself is known to act as an inhibitor of protein phosphatase 2A (PP2A), which is a major serine/threonine phosphatase. By inhibiting I2PP2A, these activators indirectly boost PP2A activity, which in turn can affect the phosphorylation status of many cellular proteins.

The biochemical interaction between I2PP2A activators and the I2PP2A enzyme is complex and highly specific. These chemical activators bind to the I2PP2A enzyme at particular domains, which alters the conformation of the enzyme and reduces its ability to inhibit PP2A. The consequential increase in PP2A activity leads to the dephosphorylation of its numerous substrates. Since phosphorylation is a common post-translational modification that alters the function, activity, and localization of proteins, the upregulation of PP2A activity by I2PP2A activators has a widespread impact on cellular processes.
