Serine peptidase inhibitor, Kunitz type 3, commonly abbreviated as SPINT3, is a protein encoded by the SPINT3 gene in humans. As a member of the Kunitz type serine protease inhibitor family, SPINT3 plays a significant role in regulating proteolytic processes by inhibiting the activity of certain serine proteases. Serine proteases are a broad class of enzymes that cleave peptide bonds in proteins, where a serine residue plays a key role in the catalytic function of the enzyme.

The Kunitz domain is a characteristic feature of this family, named after the biochemist Moses Kunitz. This domain acts as a potent protease inhibitor, and proteins with Kunitz domains are involved in a variety of biological processes, including coagulation, fibrinolysis, inflammation, and cell migration. The inhibitors typically function by forming a tight, reversible complex with the active site of the target protease, thus preventing substrate access and enzyme activity.SPINT3's precise physiological roles are not as well characterized as other members of the Kunitz inhibitor family, such as SPINT1 and SPINT2, which have been more extensively studied. However, like other Kunitz type inhibitors, SPINT3 is likely to be involved in maintaining the balance of proteolytic activity in tissues, especially in pathways related to inflammation and tissue remodeling.
