GS188 Activators function primarily by enhancing the intracellular levels of cyclic adenosine monophosphate (cAMP), a pivotal second messenger in many cellular processes. Forskolin, PGE2, Isoprenaline, Epinephrine, Dopamine, SKF38393, and BRL 37344 increase intracellular cAMP levels by activating adenylate cyclase or specific receptors linked to adenylate cyclase. On the other hand, IBMX, Rolipram, and ZM241385 raise intracellular cAMP levels by inhibiting phosphodiesterase or the A2A adenosine receptor, which normally decreases cAMP levels. The increased cAMP levels stimulate protein kinase A (PKA), which can then activate GS188 by phosphorylating it.
The activation of GS188 by PKA is a vital step in the functional activation of GS188. An increased intracellular cAMP level leads to the activation of PKA. Once activated, PKA can phosphorylate and activate GS188, thereby enhancing its functional activity. 8-Bromo-cAMP directly activates PKA, thus bypassing the need for increased cAMP levels. H-89, in contrast, functions by inhibiting PKA. This inhibition can reduce the negative feedback on GS188, thereby enhancing its activity. Therefore, the functional activation of GS188 can be achieved through various biochemical strategies, either by increasing cAMP levels or directly stimulating PKA.
