Grx2 Inhibitors

Glutaredoxin 2 (Grx2) belongs to a family of small redox proteins that have a significant role in maintaining cellular redox balance. This group of compounds, which we're referring to as Grx2 inhibitors, can influence the functionality of Grx2, either directly or by impacting the broader cellular redox environment.

Auranofin is known to inhibit thioredoxin reductase, a related protein that collaborates with Grx2 in managing cellular redox. By inhibiting thioredoxin reductase, auranofin can indirectly influence the activity of Grx2. BCNU, or carmustine, is an alkylating agent that can modify cysteine residues, and this modification can affect Grx2 activity. Curcumin, a natural compound found in turmeric, can impact the cellular redox balance, thus potentially affecting Grx2 activity. Arsenic trioxide, can effect various redox proteins and subsequently Grx2 functionality. DPI is an inhibitor of flavoproteins and might adjust the redox balance in a way that affects Grx2. Ebselen mimics glutathione peroxidase activity and could impact Grx2 by reshaping the redox landscape of the cell. NEM and Diamide, which modify and oxidize cysteine residues respectively, exert direct effects on thiol-redox proteins like Grx2. 2-AAPA, an inhibitor of glutathione peroxidase, can again reshape the cellular redox state, indirectly modulating Grx2's functionality.
