GAGE Activators

GAGE activators comprise a series of chemical compounds that enhance the immune system's recognition and response to GAGE antigens, commonly expressed in various cancer cells. Compounds like Forskolin and Resiquimod increase intracellular cAMP and activate the TLR7/8 pathway, respectively, which are mechanisms that can augment the presentation of GAGE peptides on MHC molecules, crucial for T cell-mediated immunity. Similarly, Imiquimod and Poly I:C, through TLR7 and TLR3 activation, respectively, can trigger cytokine production and immune cell maturation, thereby potentially enhancing the anti-tumor response to cells expressing GAGE. The use of CpG Oligodeoxynucleotide, which targets TLR9, further supports this immune activation by enhancing the presentation and recognition of GAGE antigens, while Concanavalin A can indirectly enhance immune response through T cell activation.

On the other hand, the introduction of Bacillus Calmette-Guerin (BCG) into the system is known to nonspecifically boost immune surveillance, which could result in increased detection and response to GAGE antigens
