FXRβ Inhibitors

FXRβ inhibitors are characterized by their capacity to modulate bile acid synthesis, lipid metabolism, and other pathways related to FXRβ's biological functions. This class comprises molecules that either directly affect FXRβ's activity or target parallel pathways, leading to an indirect inhibition of FXRβ. Notable representatives like Obeticholic Acid and GW4064 act as FXRα agonists. They can interfere with FXRβ activity by enhancing the FXRα pathway, which often competes for the same sets of cofactors or target genes. In this way, FXRβ's influence on gene transcription can be reduced.

Another interesting category within this class is represented by molecules like Rifampicin and Cyclosporine A, which operate by modulating the activities of other nuclear receptors or signaling proteins that are critical for FXRβ activation. For instance, Rifampicin activates PXR, which is known to cross-talk with FXRβ, thus indirectly dampening FXRβ's activity. Similarly, Cyclosporine A disrupts cytoplasmic binding proteins necessary for FXRβ's activation.
