FAS Inhibitors

FAS, also known as CD95 or apoptosis antigen 1 (APO-1), is a cell surface receptor belonging to the tumor necrosis factor (TNF) receptor superfamily. It plays a pivotal role in the regulation of programmed cell death, a process known as apoptosis. The FAS receptor, upon binding with its natural ligand, FAS ligand (FASL), initiates a cascade of intracellular events that culminate in apoptosis. This receptor-ligand interaction is a crucial regulatory mechanism, ensuring cellular homeostasis by removing cells that are damaged, infected, or no longer needed. The downstream signaling of the FAS receptor involves the recruitment of several adaptor proteins and the formation of a death-inducing signaling complex (DISC), ultimately leading to the activation of caspases, the enzymes responsible for carrying out the apoptotic process.

FAS inhibitors are chemical compounds that modulate the activity of the FAS receptor, specifically aiming to hinder or prevent its ability to induce apoptosis. These inhibitors can operate at various levels, such as preventing the binding of FASL to the FAS receptor, blocking the recruitment or action of adaptor proteins, or directly targeting the downstream signaling molecules in the FAS-induced apoptotic pathway.
