Chemical activators of FAM21B can engage various cellular signaling pathways, leading to the activation of this protein through post-translational modifications such as phosphorylation. Phorbol 12-myristate 13-acetate (PMA) is a potent activator of protein kinase C (PKC) and can phosphorylate FAM21B, which is a key step in its activation. Similarly, forskolin raises cyclic AMP (cAMP) levels, thereby activating protein kinase A (PKA), another kinase that can phosphorylate and activate FAM21B. Ionomycin, by increasing intracellular calcium levels, can activate calcium-dependent kinases which are capable of phosphorylating FAM21B, resulting in its activation. Another chemical, Calyculin A, inhibits protein phosphatases which otherwise dephosphorylate proteins, thus maintaining FAM21B in a phosphorylated and active state. Zinc pyrithione activates stress response pathways that may result in the activation of kinases capable of phosphorylating FAM21B. Insulin engages the PI3K/Akt pathway to phosphorylate and activate FAM21B. The Epidermal Growth Factor (EGF) binds to its receptor and initiates a cascade of kinase activations, one of the consequences of which can be the phosphorylation and activation of FAM21B. Each of these chemicals can precisely target specific kinases or pathways to ensure the activation of FAM21B, which is essential for its role in cellular functions.
