DR4 Activators

Common DR4 Activators include, but are not limited to Fluorouracil CAS 51-21-8, Doxorubicin CAS 23214-92-8, Etoposide (VP-16) CAS 33419-42-0, Taxol CAS 33069-62-4 and Cisplatin CAS 15663-27-1.

DR4 activators are compounds that stimulate the activity or expression of death receptor 4 (DR4), also known as TNFRSF10A or TRAIL-R1. DR4 is a member of the tumor necrosis factor receptor superfamily, which plays pivotal roles in various cellular processes, including apoptosis, inflammation, and immune responses. These receptors are characterized by their ability to transduce cell death signals upon binding to their respective ligands, leading to the activation of downstream signaling pathways that culminate in programmed cell death or apoptosis.

DR4, specifically, is one of the receptors for the TNF-related apoptosis-inducing ligand (TRAIL). Upon binding to TRAIL, DR4 can recruit and interact with adaptor proteins such as FADD (Fas-associated death domain), leading to the activation of caspases, a family of protease enzymes that execute the apoptotic program in cells. DR4 activators, by enhancing the receptor's activity or expression, can potentiate the apoptotic response upon TRAIL binding, leading to increased cell death in susceptible cell populations. Understanding the mechanisms and implications of DR4 activation provides profound insights into the intricate regulatory networks that govern cellular fate decisions, particularly the balance between survival and death. Exploring the realm of DR4 activators underscores the importance of death receptors in maintaining tissue homeostasis and their roles in various physiological and pathological contexts. The nuanced modulation of DR4 activity offers a window into the broader landscape of apoptosis regulation, emphasizing the delicate interplay of signals that orchestrate cell life and demise.
