CPSF Activators

CPSF Activators are diverse chemical compounds that indirectly enhance the functional activity of Cleavage and Polyadenylation Specificity Factor (CPSF) through modulation of cellular signaling pathways or the RNA processing environment. Forskolin, by increasing intracellular cAMP levels, indirectly augments the functional role of CPSF in mRNA 3'-end processing through the activation of PKA, which can phosphorylate substrates that influence CPSF activity. Similarly, Rolipram and Vinpocetine, which inhibit PDE4 and PDE1 respectively, and Caffeine, a general phosphodiesterase inhibitor, all lead to increased cAMP levels and thus potentiate CPSF activity via PKA signaling. Zaprinast and IBMX, by inhibiting PDE5 and various phosphodiesterases respectively, also raise cAMP levels, further enhancing CPSF activity through PKA-mediated phosphorylation. The cAMP analog 8-Br-cAMP directly activates PKA, promoting phosphorylation events that can modulate CPSF's role in mRNA processing. Spermidine, by promoting RNA binding, and S-Adenosylmethionine, through methylation of RNA substrates, improve CPSF's RNA-processing efficiency.

Dimethyl Sulfoxide (DMSO) facilitates the delivery of activators or substrates to CPSF, thereby indirectly increasing its activity, whereas Olomoucine, a cyclin-dependent kinase inhibitor, may enhance CPSF function by modulating the cell cycle and thus influencing CPSF's availability. DRB, on the other hand, by inhibiting RNA polymerase II, can lead to changes in mRNA synthesis and processing dynamics, potentially influencing CPSF's role in the nucleus. Collectively, these CPSF Activators, through targeted effects on cellular signaling and RNA metabolism, facilitate the enhancement of CPSF-mediated functions in mRNAI.
