cathepsin Z Inhibitors

Cathepsin Z inhibitors are primarily compounds that target cysteine proteases, a category of enzymes to which Cathepsin Z belongs. These inhibitors are often broad-spectrum, affecting multiple members of the cathepsin family, or are designed for closely related cathepsins but may show cross-reactivity with Cathepsin Z. E-64 is a well-known broad-spectrum cysteine protease inhibitor with proven efficacy against Cathepsin Z. Z-Phe-Tyr(tBu)-diazomethylketone is another potent inhibitor that has been shown to inhibit various cysteine proteases and is likely effective against Cathepsin Z. CA-074, while preferentially targeting Cathepsin B, may also inhibit Cathepsin Z due to the similarities in their active sites. Leupeptin and K11777 are broad-spectrum inhibitors that affect several cysteine proteases, potentially including Cathepsin Z. L-trans-Epoxysuccinyl-leucylamido(4-guanidino)butane (E-64) specifically targets cysteine proteases and is likely to inhibit Cathepsin Z. MDL28170, primarily a calpain inhibitor, also exhibits inhibitory activity against other cysteine proteases, suggesting a potential effect on Cathepsin Z. VBY-999, although selective for cathepsins L and S, might exhibit some level of inhibition towards Cathepsin Z due to the structural similarities among cathepsins.
