C9orf78 Activators

Forskolin, through the elevation of cAMP, activates protein kinase A (PKA), which then phosphorylates target proteins, thereby modulating their activity. Ionomycin and A23187, both calcium ionophores, increase the intracellular concentration of calcium, a critical secondary messenger that activates a suite of calcium-dependent proteins and kinases. Such activation can cascade down to affect numerous proteins within the cell.

Further modulation is seen with IBMX and PMA, the former raising levels of cAMP and cGMP by inhibiting phosphodiesterases, and the latter activating protein kinase C, a key player in phosphorylation signaling. Inhibitors of specific kinases, like genistein for tyrosine kinases, LY294002 for PI3K, PD98059 for MEK, and SB203580 for p38 MAP kinase, impact phosphorylation cascades, leading to altered protein activities. These inhibitors function by preventing the inactivation of their target proteins or by maintaining them in an activated state. U73122 disrupts phospholipase C activity, which is pivotal in generating diacylglycerol and inositol trisphosphate, secondary messengers involved in the activation of proteins. KN-93 inhibits Ca2+/calmodulin-dependent protein kinase II, consequently affecting proteins regulated by calcium signaling. Rapamycin's inhibition of mTOR, a central regulatory kinase, can have widespread effects on the cell, including the activity of downstream proteins.
