C8A Activators

Chemical activators of C8A can initiate a cascade of intracellular events leading to its functional activation. Calcium ionophore A23187 serves as a facilitator for calcium influx, a pivotal second messenger in numerous signaling pathways, including those that C8A is involved in. This influx of calcium can activate various calcium-dependent enzymes and proteins that are essential for the activation of C8A. Similarly, the compound ionomycin also functions by elevating intracellular calcium levels, thus triggering the activation of C8A through calcium-mediated signaling processes. Another chemical, Phorbol 12-myristate 13-acetate (PMA), is known to directly stimulate protein kinase C (PKC), which phosphorylates a spectrum of target proteins within the same pathway as C8A, thereby promoting its activation.

Continuing the theme, forskolin works to increase the concentration of cAMP within the cell, a critical secondary messenger that activates protein kinase A (PKA). PKA can phosphorylate and thereby activate C8A. Ouabain, through its inhibition of Na+/K+-ATPase, indirectly raises the levels of intracellular calcium, which can then activate pathways involving C8A. Thapsigargin, by inhibiting SERCA, also leads to increased cytosolic calcium that contributes to C8A activation. The use of Dibutyryl cyclic AMP (db-cAMP) similarly elevates cAMP levels, activating PKA and potentially leading to the phosphorylation and activation of C8A. Bryostatin 1, although complex in its interactions, modulates PKC activity, which can have a downstream effect on the activation of C8A. Okadaic acid, a potent inhibitor of protein phosphatases PP1 and PP2A, maintains protein phosphorylation states, which may include the phosphorylated state of C8A necessary for its activation. Prostaglandin E2 (PGE2) activates G-protein coupled receptors (GPCRs), initiating signaling cascades that activate C8A. Hydrogen peroxide (H2O2) triggers oxidative stress responses, activating redox-sensitive signaling pathways that can lead to the activation of C8A. Lastly, nitric oxide donors such as S-Nitroso-N-acetylpenicillamine (SNAP) release nitric oxide, which activates guanylate cyclase, and this activation can cascade down to activate C8A.
