ADCK5, or AarF domain-containing kinase 5, is an atypical member of the protein kinase family, encoded by the gene ADCK5. Unlike classical protein kinases, ADCK5 and its relatives within the ADCK (AarF domain-containing kinase) subfamily are characterized by the presence of a unique conserved domain structure and are considered orphan kinases because their specific substrates and regulatory mechanisms are not well understood.
ADCK5 lacks some of the conserved residues that typically define active protein kinases, which suggests that it may have a divergent enzymatic activity or possibly a non-catalytic role within cells. The protein is believed to be involved in mitochondrial biology due to its subcellular localization and similarity to other mitochondrial proteins. ADCK5 is thought to contribute to the maintenance of mitochondrial health and function, potentially influencing mitochondrial membrane potential, electron transport, and energy metabolism.Although the detailed biological functions of ADCK5 are yet to be fully elucidated, it is hypothesized that it may play a role in the coenzyme Q10 (CoQ10) biosynthesis pathway, similar to other members of the ADCK family. Coenzyme Q10 is a vital lipid component of the electron transport chain and acts as an antioxidant in both mitochondria and lipid membranes.
