Chemical activators of glutamate rich 6B can influence its activity through various intracellular signaling pathways. Calcium ionophore A23187 and Ionomycin both serve to elevate intracellular calcium levels, which in turn can trigger the activity of calcium-dependent protein kinases. These kinases directly phosphorylate glutamate rich 6B, leading to its functional activation. Phorbol 12-myristate 13-acetate (PMA) is known to directly stimulate protein kinase C (PKC), another kinase that phosphorylates serine and threonine residues on target proteins such as glutamate rich 6B, resulting in its activation. Forskolin, by increasing cellular cAMP, indirectly leads to the activation of protein kinase A (PKA), which can also target glutamate rich 6B for phosphorylation and subsequent activation. Similarly, dibutyryl-cAMP and 8-Bromo-cAMP, both permeable analogs of cAMP, activate PKA, which then phosphorylates and activates glutamate rich 6B. Isoproterenol, through its agonist action on beta-adrenergic receptors, increases intracellular cAMP and thereby activates PKA, leading to the activation of glutamate rich 6B.
In addition to these, Okadaic acid and Calyculin A work by inhibiting protein phosphatases PP1 and PP2A, which normally dephosphorylate proteins. By inhibiting these phosphatases, they ensure that glutamate rich 6B remains phosphorylated and thus in an active state. Anisomycin activates the MAPK/ERK pathway, a cascade of kinases that culminates in the phosphorylation and activation of various proteins, including glutamate rich 6B. Epidermal Growth Factor (EGF) also activates its receptor and subsequently the MAPK/ERK pathway, resulting in the activation of glutamate rich 6B. Lastly, Spermine, a polyamine, can modulate ion channel and kinase activity, which may lead to the phosphorylation and activation of glutamate rich 6B.
