Chemical activators of FAM227B can initiate a cascade of events within cells that lead to the protein's activation. Forskolin operates by directly stimulating adenylate cyclase, which catalyzes the conversion of ATP to cyclic AMP (cAMP). An increase in cAMP levels triggers the activation of protein kinase A (PKA), a crucial enzyme that can then phosphorylate and activate FAM227B. Similarly, IBMX and Rolipram, as inhibitors of phosphodiesterases, prevent the degradation of cAMP, thereby sustaining the activation state of PKA which, in turn, acts on FAM227B. The PDE4 inhibitor shares a comparable mechanism, albeit with a more targeted inhibition of the phosphodiesterase 4 subtype, resulting in increased cAMP levels and subsequent PKA-mediated FAM227B activation.
Moreover, adrenergic compounds like Epinephrine and Isoproterenol also lead to heightened cAMP levels by stimulating adrenergic receptors that activate adenylate cyclase. The ensuing PKA activation can phosphorylate FAM227B. Anisomycin, although primarily known as a protein synthesis inhibitor, can activate stress-activated protein kinases that may also target and phosphorylate FAM227B. Additionally, PMA, through the activation of protein kinase C (PKC), and Ionomycin, by increasing intracellular calcium and subsequently activating PKC, can initiate phosphorylation events leading to FAM227B activation. Bay K8644's selective activation of L-type calcium channels further contributes to the increase in intracellular calcium, which activates calcium-dependent kinases such as PKC, again leading to the activation of FAM227B. Dibutyryl-cAMP, a cAMP analog, bypasses cellular receptors and directly activates cAMP-dependent PKA, promoting the phosphorylation of FAM227B. Lastly, Zaprinast inhibits PDE5, which results in an accumulation of cGMP and activates PKG, a kinase that can also phosphorylate and activate FAM227B.
