SEE ALSO...

SIRT1 Antibody (B-10) is a mouse monoclonal IgG2b kappa light chain antibody engineered to target amino acids 448-747 of the human SIRT1 protein, facilitating the detection of SIRT1 in human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) applications. SIRT1, also referred to as SIR2, SIR2L1, and SIR2alpha, is a crucial member of the sirtuin family. SIRT1 possesses a conserved catalytic core composed of a large Rossmann-fold domain and a smaller zinc-binding domain, interconnected by a flexible linker region that is essential for NAD⁺-dependent deacetylase activity. This structural configuration allows SIRT1 to interact with a variety of substrates, including histones and transcription factors, thereby regulating key cellular processes such as gene expression, DNA repair, metabolism, and aging. The precise structure of SIRT1 is vital for function, as this determines substrate specificity, enzymatic efficiency, and interactions with regulatory proteins, making SIRT1 a pivotal target for studies investigating age-related diseases, cancer, and metabolic disorders. Anti-SIRT1 antibody (B-10) is available in both non-conjugated form and multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, providing versatile options for diverse experimental setups. By enabling detailed structural and functional analysis, SIRT1 monoclonal antibody (B-10) serves as an indispensable tool for researchers aiming to unravel the complexities of SIRT1′s role in cellular physiology and pathology, ultimately contributing to the advancement of therapeutic strategies targeting this vital protein.

Hi, why would one select the B7 over the B10 "version" of this anti-body? They appear very similar? Sorry for the basic question...

Asked by: JSAB434
Thank you for your question. These two SIRT1 monoclonal antibodies are, in fact, very similar. At this time, the main difference between the two clones is that the B-7 clone (sc-74465) is confirmed to be reactive with mouse, rat and human SIRT1, while the B-10 clone (sc-74504) is only confirmed to be reactive with human SIRT1.

If using this SIRT1 mouse monoclonal antibody for western blotting, what secondary antibody should I use?

Asked by: Anna99
We recommend using our Mouse IgG kappa Binding Protein HRP, sc-516102. A complete list of available binding proteins is available on our website here: https://www.scbt.com/scbt/browse/support-products-mouse-igg-binding-proteins/_/N-ecrety
