Caspase-8 p18 Antibody (D-8) is a mouse monoclonal IgG2a kappa light chain antibody that detects the p18 subunit and precursor of caspase-8 in mouse, rat, and human samples through western blotting, immunoprecipitation, immunofluorescence, immunohistochemistry, and enzyme-linked immunosorbent assay. caspase-8 p18 (D-8) antibody is available in both conjugated and non-conjugated forms, providing flexibility for various experimental needs. caspase-8 plays a crucial role in the apoptotic pathway, acting as an initiator caspase that activates downstream effector caspases by cleaving their inactive precursors. This activation cascade is vital for programmed cell death, particularly in response to signals from tumor necrosis factor receptor superfamily members such as TNFRSF1A and TNFRSF6/FAS. caspase-8 forms a complex with the adaptor molecule FADD, known as the death-inducing signaling complex (DISC), which is essential for activation. Once activated, caspase-8 dissociates from the DISC and further propagates the apoptotic signal by activating other caspases. The heterodimeric structure of caspase-8, consisting of the p18 and p10 subunits, is predominantly expressed in peripheral blood leukocytes, thymus, liver, and spleen, highlighting importance in immune regulation. Mutations in the CASP8 gene, which encodes for caspase-8, can lead to caspase-8 deficiency disorder (CASP8D), a condition characterized by splenomegaly and impaired apoptosis of peripheral blood leukocytes, potentially resulting in immunodeficiency due to compromised activation of T lymphocytes, natural killer cells, and B lymphocytes.
