caspase-8 Antibody (4i30) is a mouse monoclonal IgG1 antibody that detects caspase-8 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Caspase-8 plays a crucial role in the apoptotic signaling pathway, acting as the most upstream protease in the cascade initiated by tumor necrosis factor receptor superfamily members, such as TNFRSF1A and TNFRSF6/FAS. Upon activation, caspase-8 cleaves and activates downstream effector caspases, leading to the execution of apoptosis, a vital process for maintaining cellular homeostasis and eliminating damaged or unwanted cells. Caspase-8 is primarily located in the cytoplasm, where caspase-8 forms a complex with the adaptor molecule FADD, creating the death-inducing signaling complex (DISC) that is essential for activation. This localization allows for rapid response to apoptotic signals, ensuring efficient regulation of cell fate in response to stress or damage. Caspase-8 is expressed in various tissues, with highest levels found in peripheral blood leukocytes, thymus, liver, and spleen, highlighting importance in immune function and homeostasis. Defects in the CASP8 gene, which encodes for caspase-8, can lead to caspase-8 deficiency disorder (CASP8D), characterized by splenomegaly and impaired apoptosis of peripheral blood leukocytes, potentially resulting in immunodeficiency due to compromised activation of T lymphocytes, natural killer cells, and B lymphocytes.
