Caspase-8 Antibody (3C121) is a mouse monoclonal IgG1 antibody that detects caspase-8 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Caspase-8 plays a crucial role in the apoptotic signaling pathway, acting as the most upstream protease in the activation cascade triggered by tumor necrosis factor receptor superfamily member 1A (TNFRSF1A) and Fas (TNFRSF6). Upon binding to the adaptor molecule FADD, caspase-8 forms the death-inducing signaling complex (DISC), which is essential for its activation. The active caspase-8, a heterodimer composed of p18 and p10 subunits, is then released from the DISC to activate downstream effector caspases, thereby propagating the apoptotic signal. This process is vital for maintaining cellular homeostasis and eliminating damaged or unwanted cells, and defects in the CASP8 gene can lead to caspase-8 deficiency disorder (CASP8D), which is associated with severe immunodeficiency and splenomegaly due to impaired T lymphocyte, natural killer cell, and B lymphocyte activation.
