PDLIM5 Activators

The class of chemicals categorized as PDLIM5 activators predominantly operate through the modulation of kinase activity and phosphorylation states of proteins, including PDLIM5 itself. Activators of this class facilitate the activation of protein kinase A (PKA) by increasing intracellular cAMP levels. PKA can then phosphorylate PDLIM5, thereby modulating its activity. These activators by boosting cAMP levels and subsequently promoting PKA activity, regulate the phosphorylation and consequent activity of PDLIM5.

PDLIM5 activators inhibit protein phosphatases, preserving the phosphorylation state of proteins. By inhibiting the dephosphorylation activity of these enzymes, these activators indirectly maintain the activation of PDLIM5. Similarly, a subset of PDLIM5 activators modulate PDLIM5 activity by altering the phosphorylation status of PDLIM5, but through the inhibition of PKA or PKC. Lastly, activators of this class activate PKC, leading to the phosphorylation of PDLIM5.
