Fatty Acid Synthase Activators

Fatty Acid Synthase activators comprise a range of chemicals that either directly enhance the enzymatic activity or indirectly influence the cellular pathways that regulate Fatty Acid Synthase. Insulin and IGF-1 operate through the PI3K/AKT signaling pathway to promote Fatty Acid Synthase activation, whereas glucose acts as an upstream initiator that leads to insulin secretion. Citrate serves as an allosteric activator, directly binding to Fatty Acid Synthase and increasing its activity. Acetyl-CoA and Malonyl-CoA are critical substrates for the enzyme and naturally elevate its activity when present in higher concentrations.

Thyroid hormones, like T3, bind to nuclear receptors to induce FASN transcription, subsequently increasing the levels of Fatty Acid Synthase. Similarly, sex hormones such as Estradiol, Progesterone, and Dihydrotestosterone enhance FASN gene expression, thereby contributing to the enzyme's activity. Palmitate is unique in that it can directly interact with Fatty Acid Synthase to modulate its function. Leptin, on the other hand, acts indirectly by increasing insulin sensitivity.
