ZFAND2B proteasome inhibitors, such as MG132, Bortezomib, Lactacystin, Velcade, Epoxomicin, Carfilzomib, Oprozomib, and Auranofin, affect the proteolytic activity of the proteasome, thus indirectly increasing the levels of ZFAND2B if it is subject to proteasomal degradation. Inhibition of the proteasome can lead to an accumulation of ubiquitinated proteins, which may include ZFAND2B if it is among those targeted for degradation.
Other compounds in this class target upstream components of the UPS. Pyr-41 targets the ubiquitin-activating enzyme E1, preventing the initial step of ubiquitin tagging of proteins. MLN4924 inhibits the NEDD8-activating enzyme, which is essential for the function of ubiquitin ligase enzymes that tag proteins for degradation. VLX1570 and IU1 specifically inhibit deubiquitinating enzymes, which are responsible for removing ubiquitin tags from proteins, thereby regulating the degradation process. By blocking these enzymes, the degradation of ubiquitinated proteins can be affected, which may lead to an increase in ZFAND2B levels.
