XAGE-1, standing for X Antigen family member G1, is a protein encoded by the XAGE-1 gene, which is part of a broader family of XAGE genes. These genes are categorized as cancer-testis antigens (CTAs), a class of tumor-associated antigens with normal expression largely restricted to the testis, an immune-privileged site, and aberrantly activated in various cancers. XAGE-1 expression is particularly noted in Ewing's sarcoma, lung, breast, prostate, and other solid tumors.
The biological function of XAGE-1 is not fully elucidated, but it is thought to be involved in the regulation of gene expression and possibly in the immune response to cancer cells. Its restricted expression pattern makes it an attractive target for cancer immunotherapy, as it may be recognized by the immune system as a foreign antigen when expressed in tumors, allowing for the development of XAGE-1-targeted vaccines or adoptive T-cell therapies.The aberrant expression of XAGE-1 in tumors is believed to be due to dysregulated epigenetic mechanisms that normally silence CTAs in non-germline tissues. The presence of XAGE-1 in tumors can sometimes elicit a spontaneous immune response, and its detection can serve as a biomarker for certain types of cancer.
