WDR24 activators are a collection of chemical compounds that serve to indirectly enhance the functional activity of WDR24 through their influence on a variety of cellular signaling pathways. Forskolin and 8-Bromoadenosine 3',5'-cyclic monophosphate (8-Br-cAMP) enhance WDR24's activity by elevating intracellular cAMP, subsequently activating protein kinase A (PKA). PKA can then phosphorylate proteins that either interact with or regulate WDR24, thereby enhancing its activity. Similarly, ionomycin and A-23187 increase intracellular calcium concentrations, potentially activating calcium-dependent proteins that modulate WDR24's function. Phorbol 12-myristate 13-acetate (PMA) engages protein kinase C (PKC), which may phosphorylate substrates that enhance the interaction capabilities of WDR24, while FTY720 influences sphingosine 1-phosphate receptor signaling, leading to effects that may include the enhancement of WDR24 function.
Further indirect activation of WDR24 is achieved through the inhibition of key signaling molecules by compounds such as LY294002, a PI3K inhibitor, and PD98059, a MEK inhibitor, which can lead to upregulated activity of WDR24 by disinhibiting regulatory pathways. U0126 and SB203580, both inhibitors in the MAPK signaling pathway, may shift the signaling equilibrium to favor pathways involving WDR24, thereby enhancing its activity. Thapsigargin contributes to this enhancement by disrupting calcium homeostasis, which may activate calcium-dependent proteins that interact with WDR24. Lastly, epigallocatechin gallate (EGCG), through its antioxidant properties, modulates several signaling pathways, which might lead to an indirect increase in WDR24's functional activity.
