Bryostatin 1 modulates PKC in a manner that promotes phosphorylation of WDR21, while DiC8, acting as a DAG mimic, directly activates PKC, which can lead to the phosphorylation and activation of WDR21. Calcium Ionophore A23187 elevates intracellular calcium, which in turn can activate WDR21 through the action of calcium-dependent kinases. Isoproterenol, by stimulating beta-adrenergic receptors, leads to an increase in cAMP and the consequent activation of PKA, which is another kinase that can phosphorylate WDR21. Lastly, the chemical 4β-Phorbol activates PKC, which can subsequently phosphorylate and activate WDR21, completing the array of mechanisms through which these chemicals can activate the protein.
