Chemical activators of Vmn2r82 can engage different cellular pathways to initiate its activation. Forskolin, for example, is known to directly stimulate adenylyl cyclase, resulting in an elevated intracellular concentration of cyclic AMP (cAMP). The increase in cAMP activates protein kinase A (PKA), which can then phosphorylate Vmn2r82, leading to its activation. Similarly, Isoproterenol, a beta-adrenergic agonist, also raises cAMP levels, which further activates PKA, enabling the phosphorylation and activation of Vmn2r82. Pilocarpine and Carbachol, on the other hand, engage muscarinic acetylcholine receptors and stimulate phospholipase C. This enzyme increases the production of diacylglycerol (DAG) and inositol trisphosphate (IP3), which trigger the signaling pathways to activate protein kinase C (PKC). PKC then phosphorylates Vmn2r82, enabling its activation.
Nicotine activates nicotinic acetylcholine receptors, resulting in an influx of calcium ions. The elevated calcium levels activate calcium/calmodulin-dependent protein kinases (CaMK), which can phosphorylate Vmn2r82. Capsaicin directly binds to and activates transient receptor potential vanilloid 1 (TRPV1) channels, causing calcium influx that activates calcium-dependent kinases capable of phosphorylating Vmn2r82. Kainic Acid and Glutamate, acting on their respective receptors, similarly cause an increase in intracellular calcium, which subsequently activates PKC and CaMK, leading to the phosphorylation and activation of Vmn2r82. Sodium Fluoride and Aluminum Chloride both engage G-protein coupled pathways that can lead to the activation of PKA or PKC, with both kinases being able to phosphorylate Vmn2r82. Lastly, Ionomycin functions as a calcium ionophore, augmenting intracellular calcium which activates CaMK, and Phorbol 12-myristate 13-acetate (PMA) is a potent activator of PKC, both of which can phosphorylate and activate Vmn2r82 through their respective pathways.
