Similarly, Sildenafil, known for its action as a phosphodiesterase type 5 inhibitor, prevents the breakdown of cyclic GMP (cGMP), which in turn activates protein kinase G (PKG). PKG then can also phosphorylate and activate V1RE6. Ionomycin, a calcium ionophore, dramatically increases intracellular calcium levels, which activates calcium-dependent kinases capable of phosphorylating V1RE6, thus activating the protein. Another compound, Capsaicin, engages with transient receptor potential vanilloid 1 (TRPV1) receptors, causing calcium influx into the cell and subsequent activation of downstream kinases that phosphorylate and activate V1RE6.
Nicotine interacts with nicotinic acetylcholine receptors, leading to an increase in intracellular calcium and the activation of kinases that target V1RE6 for phosphorylation and activation. Glutamate, the principal excitatory neurotransmitter in the brain, activates its receptors, which in turn initiate intracellular signaling events culminating in the activation of kinases that phosphorylate V1RE6. Extracellular ATP, through its action on purinergic receptors, can also activate kinases that phosphorylate and activate V1RE6. Phorbol 12-myristate, known for its ability to activate protein kinase C (PKC), directly phosphorylates and activates V1RE6. Epidermal Growth Factor (EGF) binds to its receptor, triggering the MAPK/ERK pathway, which involves a cascade of phosphorylation events leading to the activation of V1RE6. Histamine, through its action on histamine receptors, can increase intracellular calcium or cAMP, thereby activating kinases like PKC or PKA that lead to the phosphorylation and activation of V1RE6. Caffeine, by inhibiting phosphodiesterase enzymes, maintains elevated levels of cAMP within the cell, resulting in the activation of PKA, which then activates V1RE6 through phosphorylation.
