Chemical activators of Tsx offer a range of pathways through which this protein can be functionally activated. Forskolin, for instance, directly targets adenylyl cyclase to increase intracellular cAMP levels, which in turn activates protein kinase A (PKA). Activated PKA is capable of phosphorylating Tsx, leading to its activation. Similarly, Ionomycin, by elevating intracellular calcium levels, can activate calcium-dependent protein kinases that may target Tsx for phosphorylation and subsequent activation. The chemical Phorbol 12-myristate 13-acetate (PMA) serves as an activator for protein kinase C (PKC), which is known to phosphorylate a wide range of proteins, including Tsx, thus activating it. Moreover, inhibition of protein phosphatases by Okadaic Acid and Calyculin A maintains Tsx in a phosphorylated, and hence active, state by preventing dephosphorylation.
Anisomycin engages with stress-activated protein kinases (SAPKs), such as JNK and p38 MAP kinase, which can phosphorylate Tsx, thereby facilitating its activation. Epigallocatechin gallate (EGCG) functions in a more nuanced manner, possibly activating kinases that can phosphorylate and activate Tsx through a complex signaling network. The lipid molecule Sphingosine can be metabolized into sphingosine-1-phosphate (S1P), which, through its receptors, activates downstream kinases that may further activate Tsx. The compound H-89, while known as a PKA inhibitor, can paradoxically activate other kinases through off-target effects that may lead to Tsx activation. Bisindolylmaleimide I, although primarily a PKC inhibitor, may activate alternative pathways, resulting in the phosphorylation and activation of Tsx. Thapsigargin disrupts calcium homeostasis and enhances the activity of calcium-dependent kinases, which can contribute to the phosphorylation and activation of Tsx. Lastly, Dibutyryl-cAMP, a stable analog of cAMP, bypasses upstream receptors and directly activates PKA, which is a key kinase in the phosphorylation and activation of Tsx.
