Nuclear Receptor Corepressor 2 (NCOR2), also known as Silencing Mediator for Retinoic acid and Thyroid hormone receptors (SMRT), is a crucial protein in the human body encoded by the NCOR2 gene. It plays a significant role in gene expression regulation, acting as a transcriptional corepressor for various nuclear receptors.NCOR2 operates by forming complexes with histone deacetylases (HDACs) and other nuclear receptors. These complexes alter chromatin structure through histone deacetylation, a process that generally represses gene transcription. This allows NCOR2 to modulate the activity of many genes indirectly.

This protein's functionality is not limited to a single role. It interacts with a host of receptors, including thyroid hormone receptors (TRs), retinoic acid receptors (RARs), and peroxisome proliferator-activated receptors (PPARs), thus influencing a broad range of biological processes. These include lipid metabolism, cell differentiation, and circadian rhythms.NCOR2 also has a significant role in embryonic development. It is involved in the process of organogenesis, where it helps regulate the development of organs and tissues. Disruptions in NCOR2 activity can lead to developmental disorders.In the context of disease, aberrant functioning of NCOR2 has been linked to several types of cancer, including breast and prostate cancer. Altered expression of NCOR2 can affect the normal balance of gene activation and repression, leading to abnormal cellular activity.
