TMPRSS3 Activators

TMPRSS3 activators encompass a variety of compounds that can indirectly enhance the protease's functional activity through intricate biochemical pathways. These activators are not direct stimulants of TMPRSS3 but operate by modulating the proteolytic environment in which TMPRSS3 functions. Serine protease inhibitors such as Camostat mesylate and Gabexate mesylate exert their effects by inhibiting TMPRSS3's protease counterparts, which can result in a compensatory upregulation of TMPRSS3's activity to maintain homeostasis in proteolysis. Similarly, Nafamostat mesylate and AEBSF hydrochloride contribute to the enhancement of TMPRSS3 by blocking competitive serine proteases, thereby increasing the demand for TMPRSS3-mediated cleavage of substrates.

Indirect TMPRSS3 activators also include agents that target proteases of different classes. E-64, a cysteine protease inhibitor, and Pepstatin A, which targets aspartic proteases, can lead to an augmentation of TMPRSS3 activity by shifting the proteolytic balance towards serine proteases. Other compounds like Bromhexine hydrochloride, a mucolytic agent, modulate the protease-antiprotease balance in specific tissues, resultingin an indirect elevation of TMPRSS3's activity in those environments. Inhibitors such as 3,4-Dichloroisocoumarin and Phenylmethylsulfonyl fluoride (PMSF) leverage the interconnected nature of proteolytic regulation, where the suppression of one or more protease activities can inadvertently necessitate the upregulation of alternative proteolytic pathways, including those mediated by TMPRSS3. This interplay ensures that vital proteolytic functions continue unabated, with TMPRSS3 activity potentially being upregulated to fill the void left by inhibited proteases. Moreover, natural inhibitors like Soybean trypsin inhibitor and synthetic compounds such as Aprotinin and Leupeptin hemisulfate contribute to this dynamic by specifically targeting proteases that share substrates or regulatory pathways with TMPRSS3, thereby indirectly enhancing TMPRSS3's role in protein activation through proteolytic cleavage.
