Chemical inhibitors of TMEM151A can achieve functional inhibition through various intracellular signaling pathways. W-7 hydrochloride, for instance, disrupts calcium signaling by antagonizing calmodulin, a protein that interacts with calcium ions and modulates various calcium-dependent pathways. Since calcium signaling is integral to numerous cellular functions, the antagonistic action of W-7 hydrochloride on calmodulin can impair the necessary calcium signaling required for TMEM151A's function. Similarly, Genistein, a tyrosine kinase inhibitor, inhibits the phosphorylation of proteins within the signaling pathways in which TMEM151A is involved. By inhibiting these kinases, Genistein prevents the activation of certain cellular processes that are essential for the functional activity of TMEM151A.
Additional inhibitors like KT5720 and Bisindolylmaleimide I target protein kinases such as PKA and PKC, respectively. KT5720 inhibits PKA, which plays a pivotal role in phosphorylation events crucial for TMEM151A's activity. By halting PKA's action, KT5720 can diminish TMEM151A's participation in its associated signaling pathways. Bisindolylmaleimide I, on the other hand, selectively inhibits PKC. Since PKC is involved in a multitude of cellular processes, including signal transduction and membrane receptor desensitization, its inhibition by Bisindolylmaleimide I can lead to a decrease in TMEM151A activity by disrupting PKC-mediated signaling pathways. Other inhibitors such as U73122 and Gö 6983 also contribute to the inhibition of TMEM151A. U73122 inhibits phospholipase C, thereby reducing the production of diacylglycerol and inositol trisphosphate, second messengers involved in the regulation of TMEM151A. Gö 6983, inhibiting multiple PKC isoforms, further decreases TMEM151A activity by preventing necessary phosphorylation events. Substances such as ML7, LY294002, PD98059, SP600125, SB203580, and PP2 round out the list by targeting various kinases and enzymes like MLCK, PI3K, MEK1/2, JNK, p38 MAP kinase, and Src family tyrosine kinases.
