Forskolin is a direct stimulator of adenylyl cyclase, which catalyzes the conversion of ATP to cAMP. The rise in cAMP levels activates protein kinase A (PKA), which then phosphorylates OR8G5, resulting in its activation. Similarly, isoproterenol, a beta-adrenergic agonist, binds to beta-adrenergic receptors and enhances adenylyl cyclase activity, thereby increasing cAMP production. The cAMP, in turn, activates PKA, which phosphorylates and activates OR8G5. IBMX works in a slightly different manner; it is a non-selective inhibitor of phosphodiesterases, enzymes responsible for cAMP degradation. By preventing cAMP breakdown, IBMX indirectly elevates cAMP levels, thus facilitating PKA activation and subsequent phosphorylation of OR8G5.
