This surge in cAMP can activate protein kinase A (PKA), which in turn can phosphorylate Olr853, leading to its activation. Similarly, PMA acts as an activator of protein kinase C (PKC), another kinase that can phosphorylate Olr853, assuming Olr853 is a substrate for PKC. The activation of PKC by PMA can thus facilitate the functional activation of Olr853 through phosphorylation.
Further, ionomycin, by elevating intracellular calcium levels, can activate calcium/calmodulin-dependent protein kinases (CaMKs), which may phosphorylate Olr853, enabling its activation within the calcium signaling pathway. BAY K8644, as an L-type calcium channel agonist, achieves a similar outcome by increasing intracellular calcium levels, which could lead to activation of kinases that target Olr853. Thapsigargin and Ouabain disrupt calcium homeostasis and ion gradients, respectively, creating conditions that can activate signaling cascades involving Olr853. Lastly, inhibitors of protein phosphatases like H-89, Okadaic Acid, and Calyculin A can lead to a reduction in the dephosphorylation of proteins, thereby maintaining kinases in an active state that can continue to phosphorylate Olr853, ensuring its continued activation.
