Chemical activators of Olr733 can initiate a cascade of intracellular events leading to its activation. Forskolin, for example, directly stimulates adenylyl cyclase, thereby increasing cyclic AMP (cAMP) levels within the cell. The rise in cAMP activates protein kinase A (PKA), which can phosphorylate various substrates including Olr733, leading to its activation. Similarly, Phorbol 12-myristate 13-acetate (PMA) activates protein kinase C (PKC), which is known to phosphorylate a wide range of proteins. The activation of PKC by PMA can thus result in the phosphorylation and subsequent activation of Olr733. Ionomycin, a calcium ionophore, increases intracellular calcium levels, which can activate calmodulin-dependent kinases (CaMKs). These kinases are capable of phosphorylating Olr733, thereby activating it.
Further down the line of chemical activators, BAY K8644, an L-type calcium channel agonist, indirectly raises intracellular calcium, which in turn can activate kinases that target Olr733. Thapsigargin, by inhibiting the sarco/endoplasmic reticulum Ca²⁺-ATPase (SERCA), also causes a rise in cytosolic calcium concentration, providing another route for kinase-mediated activation of Olr733. Ouabain disrupts ion gradients by inhibiting Na⁺/K⁺-ATPase, which can lead to the activation of several cellular signaling pathways that may converge on Olr733 activation. Zinc pyrithione, by increasing intracellular zinc levels, can activate cellular kinases that phosphorylate Olr733. Veratridine's action on voltage-gated sodium channels can trigger depolarization-dependent signaling pathways, culminating in Olr733 activation. Anisomycin activates stress-activated protein kinases (SAPKs), which can then phosphorylate and activate Olr733. Okadaic Acid and Calyculin A both inhibit protein phosphatases, leading to an increase in the phosphorylated state of cellular proteins, including kinases that can activate Olr733. Lastly, H-89, although commonly known as a PKA inhibitor, can paradoxically lead to the activation of alternative kinases through compensatory cellular mechanisms, which can then activate Olr733.
