Maskin, also known as TACC3 (Transforming Acidic Coiled-Coil Containing Protein 3), is a protein that plays a crucial role in cell division and growth. This protein, encoded by the TACC3 gene, is part of the TACC family, a group of proteins characterized by a highly conserved coiled-coil region.Involved in mitotic spindle assembly, Maskin contributes to cell cycle progression by facilitating the proper alignment and separation of chromosomes during mitosis. It interacts with microtubules, the structural components of the mitotic spindle, and other proteins such as Aurora-A kinase, helping to stabilize the spindle apparatus.
Maskin is also known to regulate mRNA translation in the cytoplasm. It binds to CPEB (Cytoplasmic Polyadenylation Element Binding protein), blocking the initiation of translation by inhibiting the binding of eIF4E to the mRNA cap structure. This regulation is essential for the control of protein synthesis in a variety of cellular processes, including cell cycle progression and differentiation.Aberrations in Maskin function or expression can lead to various health issues. Overexpression of Maskin has been linked to several cancers, including breast, lung, and colorectal cancers. It's considered a potential oncogene, and its overexpression often correlates with poor prognosis. In the context of neuroscience, Maskin is implicated in the regulation of local protein synthesis in neurons, influencing synaptic plasticity, a key factor in learning and memory.
