Chemical activators of Keratin 83 include a range of compounds that can induce its functional activation through various biochemical pathways, primarily by influencing the phosphorylation state of the protein. Phorbol 12-myristate 13-acetate (PMA) and 12-O-Tetradecanoylphorbol-13-acetate (TPA) are both potent activators of protein kinase C (PKC). These activators bind to PKC, enhancing its activity, which in turn leads to the phosphorylation of Keratin 83. This phosphorylation is a post-translational modification that can alter the function and activity of Keratin 83, leading to its activation. Similarly, compounds such as Phosphatidylserine and Bryostatin 1 can also activate PKC, with Bryostatin 1 binding to the enzyme and inducing a conformational change that results in its activation, subsequently leading to the phosphorylation of Keratin 83.
On another front, Forskolin raises intracellular levels of cyclic AMP (cAMP), which activates protein kinase A (PKA). PKA can then phosphorylate Keratin 83, which is a crucial step for its activation. Compounds like Calyculin A and Okadaic Acid act by inhibiting protein phosphatases 1 and 2A, enzymes responsible for the dephosphorylation of proteins. By inhibiting these phosphatases, they indirectly maintain Keratin 83 in a phosphorylated and active state. Ionomycin and Thapsigargin elevate intracellular calcium levels, which can activate calcium-dependent kinases. These kinases, upon activation, can phosphorylate Keratin 83, thus activating it. Ionomycin does this by acting as a calcium ionophore, while Thapsigargin inhibits the sarcoplasmic/endoplasmic reticulum Ca2+ ATPase (SERCA), disrupting calcium homeostasis. Diacylglycerol (DAG) is a natural ligand for PKC and, upon binding, activates the kinase which then goes on to phosphorylate Keratin 83. A23187 (Calcimycin) similarly facilitates an increase in intracellular calcium concentration, which activates calcium-dependent kinases that can phosphorylate and activate Keratin 83. Lastly, Anisomycin is known to activate stress-activated protein kinases, which then can also target Keratin 83 for phosphorylation, leading to its activation. Each of these chemicals thus contributes to the activation of Keratin 83 through the modulation of phosphorylation pathways that are essential for its function.
