Interleukin 36 Receptor Antagonist (IL36RN) is a protein encoded by the IL36RN gene in humans. This protein is part of the interleukin-1 cytokine family, which are crucial components of the immune system. They are involved in various immune responses, inflammation, and hematopoiesis, which is the formation of blood cellular components.The IL36RN protein acts as an antagonist to the interleukin-36 receptor (IL-36R). When IL-36R is activated by binding to its agonists (such as IL-36α, IL-36β, and IL-36γ), it triggers a cascade of signaling events leading to the production of pro-inflammatory cytokines. For instance, loss-of-function mutations in IL36RN can lead to a rare skin disorder known as generalized pustular psoriasis, characterized by severe skin inflammation. Additionally, research has suggested a role for IL36RN in other inflammatory conditions, including psoriatic arthritis, Crohn's disease, and ulcerative colitis.In summary, IL36RN is a significant regulatory protein in the immune system, helping to balance immune responses by antagonizing the pro-inflammatory actions of the IL-36 receptor.
