The protein IF3, or Initiation Factor 3, is a bacterial protein that plays an essential role in the initiation of protein synthesis. In the context of bacterial translation, initiation is a highly regulated process that involves the assembly of the small 30S ribosomal subunit with the messenger RNA (mRNA) and the initiator transfer RNA (tRNA) charged with N-formylmethionine (fMet-tRNA^fMet), which is the first amino acid incorporated into nascent proteins in prokaryotes.
IF3 binds to the small 30S ribosomal subunit and has multiple critical functions. It helps to maintain the 30S subunit in a conformation that is conducive to interacting with mRNA and initiator tRNA. One of IF3's primary roles is to prevent the premature binding of the large 50S ribosomal subunit before the mRNA and initiator tRNA are properly positioned. This ensures that the assembly of the functional 70S initiation complex, comprising the 30S and 50S subunits along with the mRNA and fMet-tRNA^fMet, occurs accurately and efficiently.
Additionally, IF3 plays a role in ensuring the fidelity of translation initiation. It helps in the selection of the correct start codon on the mRNA, usually AUG, and in the discrimination against non-initiator tRNAs, thus preventing the misincorporation of amino acids at the N-terminus of the protein being synthesized. IF3 also facilitates the release of deacylated tRNA from the E site (exit site) of the ribosome, resetting it for the next round of translation initiation.
