Chemical activators of FAM75C1 can be understood through their effects on various signaling pathways and cellular processes that culminate in the protein's activation. Phorbol 12-myristate 13-acetate (PMA) is a powerful activator of protein kinase C (PKC), which phosphorylates target proteins, including FAM75C1, resulting in its activation. Forskolin, by raising intracellular cAMP levels, activates protein kinase A (PKA), which similarly can lead to the phosphorylation and activation of FAM75C1. Ionomycin, through its capacity to increase intracellular calcium concentrations, activates calcium-dependent kinases that then target proteins like FAM75C1 for activation. Another synthetic DAG mimic, 1,2-Dioctanoyl-sn-glycerol (DiC8), also activates PKC and thus can promote the activation of FAM75C1. Inhibitors of protein phosphatases such as Okadaic Acid and Calyculin A prevent the dephosphorylation of proteins, leading to a net increase in the phosphorylated, and thus active, form of proteins including FAM75C1.
Further along these lines, Thapsigargin disrupts calcium storage, leading to activation of signaling cascades that ultimately could activate FAM75C1. Anisomycin, which activates stress-activated protein kinases, including JNK, can also facilitate activation of FAM75C1 through phosphorylation. Staurosporine, despite being a broad-spectrum kinase inhibitor, can at sub-inhibitory concentrations activate kinases that phosphorylate and activate FAM75C1. Paradoxically, BIM-1, primarily known as a PKC inhibitor, can at certain concentrations activate PKC, providing another route for FAM75C1 activation. The chemical 5-Iodotubercidin interferes with adenosine metabolism and thus raises adenosine levels, which through A2 adenosine receptor activation can enhance PKA activity and consequently the activation of FAM75C1. Lastly, Fusicoccin aids in the stabilization of interactions between 14-3-3 proteins and their targets, potentially fostering the active conformation and function of FAM75C1 through complex formation. Each of these chemicals, through their unique interactions with cellular signaling machinery, can contribute to the activation state of FAM75C1.
