FAM55A Activators

FAM55A include a variety of compounds that modulate cellular signaling pathways leading to its phosphorylation and subsequent activation. Forskolin, by directly activating adenylyl cyclase, raises cyclic AMP (cAMP) levels within the cell, thus enhancing the activity of protein kinase A (PKA). When activated, PKA can phosphorylate FAM55A. Similarly, Isoproterenol functions as a beta-adrenergic agonist, increasing cAMP and subsequently activating PKA, which then can target FAM55A. IBMX and Dibutyryl-cAMP, by raising intracellular cAMP, activate PKA, which in turn phosphorylates FAM55A. Rolipram, by inhibiting phosphodiesterase 4, and Zaprinast, by inhibiting phosphodiesterase 5, both result in increased cAMP levels, which activate PKA, with the latter also raising cGMP levels that can activate PKG, another kinase capable of phosphorylating FAM55A.

PMA activates protein kinase C (PKC), which has a broad range of target proteins and can phosphorylate FAM55A. Ionomycin, by increasing intracellular calcium levels, activates calmodulin-dependent kinase (CaMK), which may also facilitate the phosphorylation of FAM55A. Epinephrine, acting through adrenergic receptors, leads to cAMP production and PKA activation, which then targets FAM55A. Parathyroid hormone (PTH) activates the cAMP/PKA signaling pathway, possibly resulting in phosphorylation of FAM55A. Anisomycin, as a JNK activator, initiates a cascade that could include the phosphorylation of FAM55A. Lastly, Calyculin A inhibits protein phosphatases 1 and 2A, leading to an overall increase in protein phosphorylation, including that of FAM55A. Together, these chemicals employ a variety of signaling mechanisms to promote the phosphorylation and activation of FAM55A.
