APC membrane recruitment protein 2 (APC2) is a protein that plays a critical role in the regulation of cell division and signaling pathways related to cell adhesion and migration. APC2 is encoded by the gene ANKRD28 and is a part of the Wnt signaling pathway, which is pivotal in the regulation of gene expression, cellular growth, and embryonic development.As a component of the adenomatous polyposis coli (APC) protein complex, APC2 is involved in the destruction complex that regulates the levels of β-catenin, a key effector in the Wnt pathway. By influencing β-catenin stability, APC2 indirectly affects gene transcription and thereby has a hand in controlling cell proliferation and differentiation.

APC2 is also known to interact with microtubules and contribute to the stabilization of the cytoskeleton. This interaction is vital for proper mitotic spindle assembly during cell division. Moreover, the protein has been implicated in the migration and adhesion of cells by organizing cell matrix adhesions and influencing the actin cytoskeleton.The dysfunction or mutation of APC2 can lead to aberrant activation of the Wnt signaling pathway, which is often associated with the development of colorectal cancers and other types of tumors. Thus, APC2 has a tumor suppressor function, and its loss of function can disrupt the regulation of cell growth, leading to the formation of polyps and subsequent malignant transformation.
