EYA Activators

The first group of EYA activators are those that impact phosphatase activity. Tautomycin and Okadaic Acid are potent protein phosphatase inhibitors. EYA is a unique transcriptional coactivator with intrinsic tyrosine phosphatase activity. These phosphatase inhibitors could enhance EYA's activity by preventing its dephosphorylation, thus indirectly enhancing its function. This could lead to increased transcriptional activation of EYA target genes and influence downstream signaling events.

The second category of EYA activators includes compounds that modulate intracellular cAMP levels. Forskolin, Rolipram, 8-Br-cAMP, Epinephrine, and IBMX all have the potential to increase cAMP levels. EYA proteins can be phosphorylated by protein kinase A (PKA), which is activated by cAMP. Therefore, enhancing cAMP levels could potentially enhance EYA activity. H-89, a PKA inhibitor, could similarly influence EYA function by modulating cAMP-dependent pathways. Lastly, compounds like PMA, U73122, Milrinone, and Calmidazolium Chloride could influence EYA function through their effects on PKC-dependent pathways, phosphoinositide signaling, and calcium signaling, respectively.
