EYA2 Antibody (F-18) is a mouse monoclonal IgG2a kappa light chain antibody that detects EYA2 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-EYA2 antibody (F-18) is available as a non-conjugated format. EYA2, encoded by a gene located on chromosome 20q13.1, is a member of the eyes absent family, which consists of four related proteins. EYA2 plays a crucial role in developmental processes, particularly in myogenesis, where Pax3 induces EYA2 in a signaling cascade essential for muscle development. EYA2 features a highly conserved 271 amino acid domain at its carboxy-terminal, which is critical for function as a transcriptional activator, while the amino-terminal region is rich in proline, serine, and threonine residues, contributing to diverse regulatory roles. EYA2 is predominantly expressed in the cytoplasm during late developmental stages in various tissues, including extensor tendons, ligaments, cranial placodes, branchial arches, the central nervous system, and the developing eye. Six proteins facilitate EYA2′s translocation to the nucleus through homeodomain binding, allowing function in connective tissue patterning. Notably, EYA2′s carboxy-terminal interacts with Gαz and Gαi2 proteins, which can inhibit nuclear translocation, highlighting the intricate regulatory mechanisms governing EYA2′s activity in cellular processes.

I am using EYA2 (F-18): sc-100325 for immunofluorescence, what dilution of this antibody should I use?

Asked by: Dr Ninau Qelp
Thank you for your question. We recommend a starting dilution of 1:50. However, optimal antibody concentration should be determined by titration. Please find our full immunofluorescence protocol here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
