ENTHD1, or ENTH domain-containing protein 1, is a protein that in humans is encoded by the ENTHD1 gene. The name "ENTHD1" suggests that it contains an ENTH (Epsin N-Terminal Homology) domain, which is a conserved protein domain found in a number of proteins involved in intracellular trafficking, particularly in the endocytosis pathway.ENTH domains typically bind to phospholipids within the cell membrane, specifically phosphatidylinositol (4,5)-bisphosphate [PI(4,5)P2], and are involved in inducing curvature in the membrane that is needed for the formation of vesicles during endocytosis. This process is essential for cells, as it allows them to internalize various molecules and to recycle plasma membrane components.
Proteins with ENTH domains, such as ENTHD1, are implicated in the early stages of clathrin-mediated endocytosis, a well-characterized pathway through which many types of cargo, including nutrients, signaling receptors, and synaptic vesicles, are transported into the cell. The precise function of ENTHD1 in this process is not fully elucidated, but it may be involved in the recruitment of clathrin and adaptor proteins to sites of vesicle formation or in the stabilization of the membrane curvature necessary to form a vesicle.Given the importance of endocytosis in cellular function and communication, ENTHD1 could play a significant role in numerous cellular processes, including receptor downregulation, neurotransmitter release, and the immune response. Variations in the ENTHD1 gene or dysregulation of the protein's function could potentially impact these processes and might be relevant in the context of diseases where cellular trafficking is affected, such as neurodegenerative disorders or cancer.
