EDEM Activators

EDEM activators are a class of chemicals that play a crucial role in modulating the function of the endoplasmic reticulum degradation-enhancing mannosidase-like protein (EDEM). EDEM is primarily involved in the extraction and degradation of misfolded glycoproteins, curbing their accumulation in the endoplasmic reticulum (ER) and maintaining cellular protein quality control. These activators can directly or indirectly influence EDEM's function by targeting specific cellular pathways and processes.One group of EDEM activators includes compounds like puromycin and E-64. Puromycin directly activates EDEM by incorporating into growing polypeptide chains during protein synthesis, leading to translation errors and the generation of misfolded proteins. It can influence the cellular environment within the ER, allowing EDEM to efficiently carry out its role in glycoprotein quality control. Guggulsterone, an activator in this class, impacts lipid metabolism and ER stress pathways, ultimately enhancing EDEM's function in ERAD.
