DAZ3 Inhibitors

DAZ3 inhibitors predominantly target translational regulation and stress-response pathways to exert their effects. Among these, Rapamycin is noteworthy for its inhibition of mTOR, which leads to diminished phosphorylation of S6K. The reduction in S6K activity directly affects ribosome biogenesis, impacting the translational processes that DAZ3 seeks to control. Similarly, Wortmannin and LY294002 obstruct PI3K, thereby reducing Akt phosphorylation levels, which in turn influence mTOR signaling. This results in a constriction of the regulatory actions traditionally managed by DAZ3.

Moreover, Actinomycin D and 5-Azacytidine serve to modify the mRNA landscape within the cell. Actinomycin D, for instance, interferes with RNA polymerase, leading to a disruption in mRNA synthesis. This alters the pool of mRNAs that would otherwise be subject to post-transcriptional control by DAZ3. On the other hand, 5-Azacytidine changes the DNA methylation pattern, causing alterations in gene expression profiles.
