Syncytin-B is an endogenous retroviral protein encoded by the envelope (env) gene of the human endogenous retrovirus W (HERV-W) family. This fusogenic protein is expressed in the human placenta and is pivotal in the process of placentation, which is essential for proper embryonic development and successful pregnancy outcomes.The primary role of Syncytin-B is to facilitate the fusion of cytotrophoblasts into the multinucleated syncytiotrophoblast layer, a critical step in the formation of the placenta. The syncytiotrophoblast forms the outermost layer of the placental villi and serves several vital functions, including mediating nutrient and gas exchange between the mother and the fetus, secreting hormones that regulate pregnancy, and acting as a barrier to protect the fetus from pathogens.

Syncytin-B operates by binding to specific receptors on the surface of cytotrophoblast cells, triggering a cascade of events that lead to the merging of cell membranes. This fusogenic activity is facilitated by the protein's structure, which is characterized by a surface unit (SU) responsible for receptor binding and a transmembrane unit (TM) that promotes the fusion of cellular membranes.The expression of Syncytin-B is tightly regulated, as inappropriate expression can lead to pathological conditions. For instance, aberrant expression has been associated with certain pregnancy-related disorders, such as pre-eclampsia. Additionally, Syncytin-B's role in cell fusion extends beyond placentation; its expression has been detected in some cancerous tissues, suggesting a potential involvement in the pathological cell fusion events observed in cancer progression.Understanding the function and regulation of Syncytin-B is not only critical for elucidating the mechanisms underlying placental development and related pathologies but also for exploring its potential role in cancer biology and as a target for intervention.
