D54 Activators

D54 Activators can be categorized into two main types: those that increase cAMP levels and those that inhibit cAMP breakdown. The first category includes forskolin, isoproterenol, 8-Bromo-cAMP, PGE2, epinephrine, dobutamine, N6,2'-O-Dibutyryladenosine 3',5'-cyclic monophosphate, terbutaline, and salbutamol. These compounds stimulate adenylate cyclase or are cell-permeable cAMP analogs that increase the levels of cAMP in the cell. The elevated cAMP levels can activate protein kinase A (PKA), a known phosphorylator of D54. Phosphorylation by PKA can enhance the functional activity of D54, making these compounds effective D54 activators.

The second category includes rolipram and IBMX, which are phosphodiesterase inhibitors that prevent the breakdown of cAMP, leading to increased cAMP levels. Elevated cAMP levels can activate PKA, which can phosphorylate and activate D54. The unique action of H-89 dihydrochloride, an inhibitor of PKA, also indirectly enhances the activity of D54,which is known to be negatively regulated by PKA-mediated phosphorylation. Inhibition of PKA releases this negative regulation, leading to increased D54 activity.
